Inhibition of NF-kappa B can enhance Fas-mediated apoptosis in leukemia cell line HL-60

摘要: This study explored the effects of nuclear factor-kappa B (NF-κB) inhibitor Bay 11-7082 on Fas/FasL system and Fas-mediated apoptosis in cell line HL-60 cells. The mRNA and protein levels of Fas, FasL, and X-linked inhibitor of apoptosis protein (XIAP) were detected by reverse transcription-polymerase chain reaction (RT-PCR) and flow cytometry (FCM); the level of sFasL was evaluated by enzyme-linked immunosorbent assay (ELISA); and apoptosis was determined by FCM. After treatment with Bay 11-7082, the mRNA and protein levels of FasL and XIAP in HL-60 cells were significantly lower than in the controls ( <0.05), but the mRNA and protein levels of Fas and sFasL did not change significantly ( >0.05). Apoptotic rate of HL-60 cells treated with Bay 11-7082 was significantly higher than in the controls ( <0.05). Therefore, we conclude that Bay 11-7082 can enhance Fas-mediated apoptosis in HL-60 cells by downregulating FasL and XIAP levels.

Palmitoylation of GNAQ/11 is critical for tumor cell proliferation and survival in GNAQ/11-mutant uveal

摘要: More than 85% of patients with uveal melanoma (UM) carry a GNAQ or GNA11 mutation at a hotspot codon (Q209) that encodes G protein α subunit q/11 polypeptides (Gαq/11). GNAQ/11 relies on palmitoylation for membrane association and signal transduction. Despite the palmitoylation of GNAQ/11 was discovered long before, its implication in UM remains unclear. Here, results of palmitoylation-targeted mutagenesis and chemical interference approaches revealed that the loss of GNAQ/11 palmitoylation substantially affected tumor cell proliferation and survival in UM cells. Palmitoylation inhibition through the mutation of palmitoylation sites suppressed GNAQ/11Q209L-induced malignant transformation in NIH3T3 cells. Importantly, the palmitoylation-deficient oncogenic GNAQ/11 failed to rescue the cell death initiated by the knock down of endogenous GNAQ/11 oncogenes in UM cells, which are much more dependent on Gαq/11 signaling for cell survival and proliferation than other melanoma cells without GNAQ/11 mutations. Furthermore, the palmitoylation inhibitor, 2-bromopalmitate, also specifically disrupted Gαq/11 downstream signaling by interfering with the MAPK pathway and BCL2 survival pathway in GNAQ/11-mutant UM cells and showed a notable synergistic effect when applied in combination with the BCL2 inhibitor, ABT-199, in vitro. The findings validate that GNAQ/11 palmitoylation plays a critical role in UM and may serve as a promising therapeutic target for GNAQ/11-driven UM.

Effects of the SAPO-11 synthetic process on dehydration of ethanol to ethylene

摘要: The effects of the synthetic condition of SAPO-11 molecular sieves on ethanol dehydration to ethylene were studied. Product-compositions, ethanol conversion, and selectivity to ethylene of synthesized and commercial SAPO-11 molecular sieves were compared. Results are as follows: the optimal synthetic conditions for SAPO-11 molecular sieves are adding pseudoboehmite before orthophoshporic, using di-n- propylamine as the template, having a mass fraction of 40% colloidal silica as the silica source and the starting gel obtained, and running at 200°C for 48 h. From the patterns of NH -TPD, the amount of acid synthesized by SAPO-11 molecular sieves is less than that by commercial SAPO-11 molecular sieves, and has a stronger weak acid. Also, ethanol conversion and selectivity to ethylene reached 99% at 280°C on synthesized SAPO-11, lower by 20°C compared to commercial SAPO-11. For two SAPO-11 molecular sieves, the by-products in the gas phase are mainly ethane, propane, propene, isobutane, n-butane, propadiene, butylene and some higher hydrocarbons. The by-products in the liquid phase are ethyl ether and acetaldehyde.

海湾扇贝引种工程及其综合效应

摘要:

回顾了海湾扇贝(Argopecten irradians)引种的社会与学术背景、引种、产业化和种质复壮等过程,探讨了引种工程的经济、社会效益及其产生的综合效应。20世纪70年代末,为了缓解我国黄渤海海域养殖业面临滑坡的态势,我们选定并引进了美国的海湾扇贝。在实现人工繁育技术系列化和养殖过程全人工化的基础,世界上第一个海湾扇贝养殖业从此成为我国浅海养殖的支柱产业之一。引种工程在东海和南海也得以拓展且颇见成效。养殖群体的种质复壮,为该产业持续发展奠定了“种质”基础。尚未发现海湾扇贝在中国海形成自然种群,表明该项引种工程不会对本地种的种质和资源造成负影响。其生态环境效应主要体现在因产生大量生物性沉积而导致的自身污染,但与近年的栉孔扇贝大规模死亡并无直接的因果关系。该项引种工程瞄准了国家需求,产生了巨大经济、社会效益,提高了我国贝类研究在国际上的地位。海湾扇贝引种工程意义重大,前景广阔,并将产生持续、深远的综合效应。

Evaluation of the ecological status with benthic indices in the coastal system: the case of Bohai Bay

摘要: Based on biologic and environmental materials collected from coastal areas of Bohai Bay (China) in April, 2008, three biotic indices (AZTI’s Marine Biotic Index (AMBI), Shannon–Wiener Index and -statistic) were applied together to evaluate the ecological status of the sampling area. The results showed a clear spatial gradient from a worse ecological status in the near-shore areas (especially around Haihe and Jiyun River Estuaries) to a better status in the offshore areas. While all the three indices could assist decision makers in visualizing spatial changes of organic pollutants in Bohai Bay, two indices, i.e., AMBI and Shannon-Wiener index, were effective in distinguishing sites from Haihe River Estuary, Jiyun River Estuary and other area. However, -statistic can’t tell the differences between estuaries and other area. It would be explained that organic pollutants and/or other environmental stresses in Bohai Bay were not strong enough to reduce the size of macrozoobenthos, which may cause both of the abundance and biomass curves crossed. To our knowledge, this is the first time that several benthic indices were used to assess the benthic ecological status in Bohai Bay, which gave the similar results. Furthermore, there is indication that the ecological status is related to excess input of wastewater along main rivers and outlets. In a word, AMBI, Shannon–Wiener Index and -statistic could be able to assess the benthic ecological status of Bohai Bay under the organic pollutants pressure.

渤海湾是我国典型的平原淤泥质海岸,中国最大的两个人工港--天津新港和河北黄骅港,在建设过程中曾先后遇到细颗粒泥沙运移与如何减轻港口、航道回淤量等工程泥沙问题。文章以渤海湾西南部大口河沿岸海域现场观测资料为基础,通过近岸水动力、泥沙输移、岸滩沉积过程及冲淤演变等综合分析,为平原淤泥质海岸"浅水深用"的建港战略决策提供理论依据。

Dynamic mechanism for the decline of China’s energy intensity during the 11th FYP (Five-Year Plan)

摘要: From 2006 to 2009, the aggregate energy intensity of China fell by 14.38%, which means that China has made substantial progress towards its goal of achieving approximately a 20% reduction in energy intensity during the period of the 11th FYP (Five-Year Plan). This paper describes some new properties of the mechanism for the decrease in China’s energy intensity since the beginning of the 11th FYP. First, compared with the period of the 10th FYP, technical, structural, and residential energy-savings have been improved to a certain extent. Secondly, unlike the dynamic mechanism before 2000, technical energy savings have been the main driver of momentum since the beginning of the 11th FYP. Next, although structural energy savings are not the main factor, they are still a key factor for the decrease in energy intensity. Finally, residential energy savings are still a stable driver of momentum for the decline in China’s energy intensity.

WLAN 802.11/11b数据加密机制的安全分析

摘要:

在802.11标准中的加密采用WEP协议,用于提供链路层数据传输的安全保护。目前,在原有EP的基础上提出了一些改进方案,能提高WEP的安全性能,但理论上缺少严密的安全分析。笔者通过数学模型对这些解决方案以及原有WEP协议进行量化分析,推导出机制内各模块与整个安全机制间安全性能的对应函数关系,并比较了这些方案间安全性能的差异,证明这些安全机制可以提高原有WEP的安全性能,在理论上为用户提供如何构造满足所需安全性能的WLAN数据加密增强机制。

在粤港澳大湾区建设进入新阶段的背景下,为进一步提升粤港澳大湾区科技创新与新兴产业的发展能级与全球影响力,支撑我国科技创新与新兴产业的高质量发展,本文基于湾区经济在国家经济发展中的作用分析、粤港澳大湾区科技创新与新兴产业的发展现状评估和国家定位研判,在“一国两制”的独特制度框架下提出了粤港澳大湾区科技创新与新兴产业中长期发展的战略目标与重点任务。面向 2050 年,粤港澳大湾区可分“四步走”全面加速建成开放、融合、可持续的全球创新与产业高地,以科技创新发展一体化来有序推动建设“无边界”的大湾区。研究提出了重点任务建议:强化国际科技创新中心建设、提升科技对产业发展的支撑能力、培育世界级新兴产业集群、打造海内外优秀人才的聚集地、推进金融与科技深度融合、建立多元化的科创投入协同机制、探索创新与产业要素流动机制。

摘要: The impact on the environment of radionuclide release from nuclear power plants has attracted increased attention, especially after the accident at Fukushima Daiichi Nuclear Power Plant in Japan. Based on the mechanisms of adsorption/desorption at solid/liquid interfaces and a surface micromorphology model of sediments, a theoretical expression of the distribution coefficient is derived. This coefficient has significant effects on the distribution of radionuclide in seawater, suspended sediment and seabed sediment. is then used to simulate Sr transport in the sea near the Daya Bay Nuclear Power Plant. The simulation results are compared with field measurements of tidal level, current velocity, suspended sediment concentration and Sr concentrations in the same period. Overall, the simulated results agree well with the field measured data. Thus, the derived expression for is capable of interpreting realistic adsorption/desorption processes. What’s more, conclusion is drawn that about 40% Sr released by Daya Bay Nuclear Power Plant will be adsorbed by suspended sediment and 20% by seabed sediment, only about 40% Sr will remain in the sea near Daya Bay Nuclear Power Plant in South China Sea.
